Target gene experiment description: Analysis of FOXA1 ChIP-seq previously performed in LNCaP cells identified strong FOXA1 binding at an Enhancer of approximately 3.7 kb upstream of the TGFB3 gene promoter.Importantly, qRT-PCR analysis demonstrated that CRISPR-mediated Enhancer deletion in only one-fourth of the cell population was able to substantially restore the transcription of TGFB3 gene, comparing cells transfected with TGFB3-targeting gRNAs to those with control gRNAs.
