Enhancer ID: | E_02_0913 |
Species: | human |
Position : | chr7:116374406-116376406 |
Biosample name: | |
Experiment class : | Low throughput |
Enhancer type: | Enhancer |
Disease: | Melanoma |
Pubmed ID: | 24443471 |
Enhancer experiment: | 3C,ChIP |
Enhancer experiment description: | Chromosome conformation capture (3C)was performed in COLO829 cells in the presence or absence of PLX4032 using the MET +63-kb enhancer as an anchor region. We observed that BRAF inhibition strikingly increased the interaction between the MET +63-kb enhancer and the MET TSS, demonstrating that this lineage-specific enhancer undergoes inducible chromatin looping in response to oncogene withdrawal in melanoma cells. |
Target gene : | MET(AUTS9,DFNB97,HGFR,RCCP2,c-Met) |
Strong evidence: | 3C |
Less strong evidence: | ChIP |
Target gene experiment description: | Chromosome conformation capture (3C)was performed in COLO829 cells in the presence or absence of PLX4032 using the MET +63-kb Enhancer as an anchor region. We observed that BRAF inhibition strikingly increased the interaction between the MET +63-kb Enhancer and the MET TSS. |
TF name : | MITF(CMM8,COMMAD,MI,WS2,WS2A,bHLHe32) |
TF experiment: | ChIP-seq,3C |
TF experiment description: | To test the necessity of MITF for inducible chromatin looping between the MET +63-kb Enhancer and MET TSS in response to BRAF inhibition, 3C was performed in COLO829 cells in the presence or absence of PLX4032 and MITF depletion. |
Enhancer function : | This enhancer displays inducible chromatin looping with the MET promoter to up-regulate MET expression upon BRAF inhibition. |
Enhancer function experiment: | ChIP,3C |
Enhancer function experiment description: |
ChIP and 3C analysis were performed on wild-type or Enhancer-edited cells treated with PLX4032.A significant decrease in both H3K27 acetylation,a histone modification associated with active Enhancer state, and Enhancer-promoter chromatin looping was observed in Enhancer-edited cells. |
